She kinda didn't make a super smash hit in this era. It doesn't need to be a smash to be a signature hit, I mean, when its all said and done the song is going to have 200m+ views and 3 million copies sold in the US to its name alone, but aside from that, there's something about the whole Applause theme that just reminds you of Gaga, in a way. Just look at artRave videos when Applause comes everyone starts clapping during the pre chorus, its just little things and moments like that that make a signature hit for an artist. Maybe the rushed release? That's the only thing I'll remember on Applause and the beat, maybe? I like the atmosphere in artRave when Applause plays though. It's without a doubt the face of ARTPOP, and it'll possibly always be remembered for that. Anything beyond that such as being considered as a signature hit for Gaga comparable to her 4 biggest global hits will depend on how history would judge it. The song is pretty memorable and it could only be a Gaga song and its very useful in all kinds of events as well. A memorable song doesn't have to do with chart success alone.
