Many people see a moustache as a symbol of masculinity and many men strive to grow one, so her she's saying that tonight it's Gaga, a woman, who is wearing the stache and is in charge, whilst also embracing her womanhood and her s-xuality by stepping out in her underwear and heels, showing her body revolution and that a women need not hide herself in front of an audience, especially at an important event like this.
