I don't know anything about this, but I would obviously trust a fansite rather than a Twitter insider.

One thing though, from his tweets it sounded like the Lady Gaga Now guy got access to some sort of database (that lists stuff that number of tracks, release dates, release formats, etc.), and I just don't see how they would know if there will or will not be a song with pre-order from that.

Exactly. A song that just comes with pre-order without any promotion or being sent to radio wouldn't take thunder away from the movie. 

I even would just be okay with a 2nd trailer released with the pre-order which shows some more of the music being performed.

They should release a second trailer with a single/song and preorders on iTunes. If the song is good enough (which I know it will be) it will keep people interested until the movie release, IMO.
